建立了超高效液相色谱-四极杆/静电场轨道阱高分辨质谱快速筛查和确证粮谷产品中20种真菌毒素的方法。样品经乙腈(含2%(体积分数)甲酸)提取,用Captiva EMR-Lipid小柱净化,采用Thermo Hypersil Gold C18柱(100 mm×2.1 mm,1.9 μm)分离,用四极杆/静电场轨道阱高分辨质谱进行分析。在全扫描模式下以分析物的保留时间和一级母离子信息实现快速筛查,以自动触发采集的二级碎片离子信息进行确证。结果显示,目标分析物在各自的质量浓度范围内线性关系良好(相关系数r2>0.99),方法检出限为0.25~20 μg/kg,回收率为72.9%~117.8%,相对标准偏差为2.9%~15.2%(n=6)。该方法灵敏度高,结果准确、可靠,适用于粮谷产品中20种真菌毒素的快速筛查和确证。  相似文献

Mycotoxins represent a wide range of secondary, naturally occurring and practically unavoidable fungal metabolites. They contaminate various agricultural commodities like cereals, maize, peanuts, fruits, and feed at any stage in pre- or post-harvest conditions. Consumption of mycotoxin-contaminated food and feed can cause acute or chronic toxicity in human and animals. The risk that is posed to public health have prompted the need to develop methods of analysis and detection of mycotoxins in food products. Mycotoxins wide range of structural diversity, high chemical stability, and low concentrations in tested samples require robust, effective, and comprehensible detection methods. This review summarizes current methods, such as chromatographic and immunochemical techniques, as well as novel, alternative approaches like biosensors, electronic noses, or molecularly imprinted polymers that have been successfully applied in detection and identification of various mycotoxins in food commodities. In order to highlight the significance of sampling and sample treatment in the analytical process, these steps have been comprehensively described.  相似文献

An ultrahigh-performance liquid chromatography–tandem mass spectrometry method was developed and validated for the simultaneous determination of sterigmatocystin, verruculogen, enniatin A, fusarenon-X, fumonisins B1, B2, B3, aflatoxins B1, B2, G1, G2, ochratoxin A, deoxynivalenol, 3-acetyldeoxynivalenol, 5-acetyldeoxynivalenol, and zearalenone. The mycotoxins were extracted and cleaned up using a multitoxin column, separated on a C18 column, and then detected on a triple-quadrupole mass spectrometer. The limits of detection and quantification ranged within 0.2–2?µg/kg and 1–10?µg/kg, respectively. The recoveries ranged from 70.8 to 118.4%, with relative standard deviations below 15%. The method was used to analyze 80 samples obtained from Shandong Province in China. Fifty-eight samples were contaminated with 10 mycotoxins at concentrations ranging from 1.4 to 6566.1?µg/kg. Some samples exceeded the maximum limits in China and in European regulations for mycotoxins in unprocessed maize.  相似文献

依次采用磷酸盐缓冲液(PBS)和甲醇-PBS溶液提取样品,以多功能免疫亲和柱净化,采用液相色谱-串联四极杆质谱检测,可同时测定中药材中的黄曲霉毒素B1、黄曲霉毒素B2、黄曲霉毒素G1、黄曲霉毒素G2、伏马毒素B1、伏马毒素B2、T-2毒素、HT-2毒素、赭曲霉毒素A(OTA)、玉米赤霉烯酮等14种真菌毒素。优化条件下,真菌毒素的定量限(LOQ)为1~5 μg/kg, 4种中药材基质(人参、桔梗、板蓝根、麦门冬)中3个不同添加水平的平均回收率(n=6)为71.9%~99.7%,相对标准偏差(RSD)为4.8%~15.8%。该方法的检测速度快,中药材复杂基质的干扰较少,结果准确、可靠,定量限可满足国内外中药材真菌毒素相关限量的要求。  相似文献

Summary Separation and determination ofFusarium trichothecenes: nivalenol, deoxynivalenol, fusarenone and 3-acetyldeoxynivalenol, were carried out by highperformance liquid chromatography with UV detection, and methanol—water (30/70) as mobile-phase on Lichrosorb RP-18 column. Detection limits were 2, 3, 3 and 5ng per injection of nivalenol, deoxynivalenol, fusarenone and 3-acetyldeoxynivalenol, respectively. The method appears to be adequate for detection of the above trichothecenes in contaminated corn and rice as well as in cultures ofFusarium spp. A preliminary TLC purification was necessary for detection of fusarenone in corn.  相似文献

采用液相色谱-串联质谱建立了小麦中T-2、玉米赤霉烯酮(ZEN)和脱氧镰刀菌烯醇(DON)的测定方法。样品经80% 乙腈-水提取,氨基柱(500 mg,6 mL)杂质吸附模式净化,以5 mmol/L乙酸铵溶液和甲醇为流动相,ZORBAX Extend-C18柱(150 mm×2.1 mm,1.8 μm)进行色谱分离;在电喷雾正离子化模式下,多反应监测方式测定。结果表明:T-2、ZEN和DON分别在0.5~500、5~500、10~2 000 μg/L质量浓度范围内呈良好线性,相关系数分别为0.998 9、0.999 7和0.999 1。通过对空白小麦样品进行3个水平的加标回收实验,T-2、ZEN和DON的回收率分别为86%~94%、80%~101%和81%~105%,相对标准偏差(RSD)分别为2.6%~5.5%、3.6%~8.9%和2.2%~8.1%,方法检出限分别为0.5、8.0、10.0 μg/kg。该方法准确、灵敏、成本低,适用于小麦中T-2、ZEN和DON的同时分析。  相似文献

Pistachios are one of the types of tree nut fruits with the highest mycotoxin contamination, especially of aflatoxins, worldwide. This study developed a Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) method that was followed by Ultra-High Performance Liquid Chromatography combined with Time-of-Flight Mass Spectrometry (UHPLC–ToF-MS) for the determination of mycotoxins in pistachios. Different approaches to dispersive solid phase extraction as a clean-up method for high lipid matrices were evaluated. For this, classic sorbents such as C18 (octadecyl-modified silica) and PSA (primary secondary amine), and new classes of sorbents, namely EMR-Lipid (enhanced matrix removal-lipid) and Z-Sep (modified silica gel with zirconium oxide), were used. The QuEChERS method, followed by Z-Sep d-SPE clean-up, provided the best analytical performance for aflatoxins (AFB1, AFB2, AFG1 and AFG2), ochratoxin A (OTA), zearalenone (ZEA), toxin T2 (T2) and toxin HT-2 (HT2) in pistachios. The method was validated in terms of linearity, sensitivity, repeatability, interday precision and recovery; it achieved good results according to criteria imposed by Commission Regulation (EC) no. 401/2006. The method was applied to real samples and the results show that pistachios that are available in Portuguese markets are safe from mycotoxins that are of concern to human health.  相似文献

真菌毒素是由真菌在一定环境条件下产生的一类具有毒性的小分子次级代谢产物。真菌毒素种类多,毒性强,污染范围广,可经食物链直接或间接进入人体,危及人体健康。食品基质形态多样,成分复杂,而实际样品中真菌毒素含量低,难以直接对目标物进行分析,故高效的样品前处理技术能实现待测物的分离和富集,在实际样品的分析中尤为重要。该文主要综述了基于磁性纳米材料、石墨烯类材料、分子印迹材料、免疫亲和材料、适配体功能材料等新型分离介质的液相萃取技术、固相萃取技术、场辅助提取技术(磁性固相萃取、超声辅助提取、微波辅助提取)、免疫亲和柱法、QuEChERS法等前处理技术在食品中真菌毒素分析中的应用,并对其分析的发展趋势进行了展望。  相似文献
